Area of Specialization

Biochemical Engineering

Expertise

Bioprocess Engineering, Fermentation technology Enzyme technology and Bio-energy

Production and purification of enzymes:-

  • Production of enzyme: Solid State Fermentation (SSF) and Submerged fermentation (SmF).
  • Purification of enzyme: Ammonium sulphate precipitation, size chromatography and affinity chromatography
  • Biochemical assays: Quantitative estimation of carbohydrates, proteins, nucleic acids using spectrophotometer and HPLC

Techniques used for analysis:-

  • SDS PAGE, Native PAGE, Zymography techniques and Western Blotting
  • Thin layer Chromatography, Gas Chromatography (GC), Fermenter, Fourier Transform Infrared Spectroscopy (FTIR), UV-Vis Spectrophotometer and Fluorescent microscope.
  • Chemical Oxygen Demand (COD), Biological Oxygen Demand (BOD), Dissolve Oxygen (DO) and CO2 measurement.

Analysis of fermentation parameters:-

Fermentation parameters: Initial total reducing sugars, So (g/L), Maximum ethanol concentration (g/L), Maximum cell concentration (g/L), Sugar consumed (%), Specific growth rate (h-1), Ethanol yield coefficient, Yp/s (g/g), Biomass yield coefficient ,Yx/s (g/g), Ethanol productivity, Qp (g/L/h), Growth rate, Qx (g cells/L/h) and Max. sugars consumption rate, Qs (g /L/ h) was calculated.

Data Analysis:-

Statistical analysis of experimental data was conducted using the PROC GLM functionality in SAS, version 9.1.3 (SAS Institute Inc., Cary, NC), Minitab 15 (Minitab Inc., USA) and Design Expert software. Both the linear and quadratic effects and the possible interactions of the variables were calculated.
